Research Abstract

<研究の目的と進め方>
我々はこれまでマイクロアレイを用いたDNAメチル化の網羅的解析法であるMIAMI法を用いて肺癌と正常肺との比較し、癌で脱メチル化されている配列の近傍において有意に多い配列が、はmicroRNA(miRNA)のひとつで3'側に末端の配列と一致していることをみいだした。ところで哺乳類でRNAinterference(RNAi)機構と転写の関係はあまりわかっていないが、最近、プロモーター配列に対する21merの2重鎖RNA(dsRNA)を導入することにより転写活性化がおこるdsRNA-induced gene activation(RNAa)という現象が報告されている。そこで我々はmiRNAの転写制御への関与の検証をおこなう。さらにMIAMI法による解析の対象を癌以外にも広げてデータベース化し、同様のモチーフ解析、機能の検証を進め、見つかってきた転写制御miRNAの共通性を探る。
<2009年度の成果>
転写開始点近傍にmiRNAと相補性をもつ遺伝子を数え上げ、転写開始点近傍にそのターゲットが有意に濃縮しているかどうかを調べた。その結果、転写開始点上流でmiRNAとマッチする配列がランダムな配列と比べて有意に多いことがわかった。一方センス鎖とアンチセンス鎖との間でターゲットの出現頻度の差はなかった。
またこれらのmiRNAの中でmiR-663が最もそのターゲットが転写開始点近傍に多いことがわかった。miR-663は乳がんで発現が減少していることが報告されているが(Lehmann U et al.J Pathol.2008 ; 214(1): 17-24.)、ターゲットとなる転写開始点近傍領域をもつ遺伝子のオントロジー解析をおこなったところ、興味深いことにシグナル伝達やキナーゼといった遺伝子が有意に多かった。
