疾患関連変異探索のための遺伝統計学的解析法の開発とデング出血熱関連解析への応用

Research Abstract

(1)ゲノムワイド関連解析または候補遺伝子アプローチによって、あるSNPマーカーにおいて有意な関連が検出された場合に、そのSNPマーカーおよび周辺の連鎖不平衡情報をHapMapデータベースより取得する。実際にタイピングしたSNPの情報とHapMapデータベースの情報とを組み合わせることで、実際にはタイピングしていないSNPでの関連検定の結果を予測するアルゴリズムを開発した。
(2)系統樹理論に基づき、ハプロタイプブロック中に存在するハプロタイプの中から、系統発生的に感受性変異を共有しうるハプロタイプの組合せをまず検出する。ハプロタイプの系統樹を作成し、未知のSNPを共有しうるハプロタイプの組合せをもとめ、それらのハプロタイプの組合せについてのみ検定を行う。検定するハプロタイプの組合せを絞ることで、分子系統進化上起こり得ない組合せから生じる偽陽性率を計算し、当該手法の有効性を評価した。
(3)SNP頻度情報などをデータベースから抽出し、有害な変異の頻度スペクトラムをもとめる。有害変異の対立遺伝子頻度は中立なものより低いが、疾患と関連している可能性は高いので、得られた頻度スペクトラムをもとに、有害変異のスクリーニングに必要な患者検体数、関連解析のサンプル数などを算出するアルゴリズムを開発した。
(4)トール様受容体遺伝子(TLR)ファミリー(TLR1、TLR2、TLR3、TLR4、TLR7、TLR8、TLR9)、各種サイトカイン(IL3、IL4、IL13)を候補遺伝子群として、デング出血熱関連解析を行った。
