Research Abstract

1) ロースワイド関連解析:III型代謝型グルタミン酸受容体遺伝子GRM4とGRM7について解析を行った。まずGRM4, GRM7について8個、および43個のSNPを選択し、100ペアのサンプルを用いて単点およびハプロタイプ関連解析を行った。GRM4については関連が認められなかったが、GRM7の2つのSNPについてハプロタイプで関連が認められた。またこの有意差はFDRによる多重検定補正後も消失しなかった。そこでこの2つのSNPにつきサンプルを追加して(合計404のケースと420のコントロールサンプル)解析を行ったところ、有意差が認められた。以上の結果から、GRM4については日本人の統合失調症発症への主要な関与はないが、GRM7は疾患感受性遺伝子と考えられた。
2) ゲノムワイド関連解析:約3万個のマイクロサテライトマーカーを用いたゲノムワイドの関連解析で、有意差のあるマーカーを基点としたSNPによる解析を行い、31個のSNPを選択した。これらのSNPにつき、独立の2,450ペアのサンプルにつき解析を行った。SLC23A3とC2orf24遺伝子間領域の1個のSNPで有意差が見られ、さらに周辺SNPのタイピングを全サンプル約2,900ペアで行った。その結果、先に有意差が見いだされたSNPとともに、C2orf24の3'フランキング領域、およびそのコード領域内のSNPにも有意差が見られ、C2orf24が疾患感受性遺伝子であることが示唆された。
3) 遺伝子改変マウスの解析:既に関連を報告している代謝型グルタミン酸受容体3型遺伝子GRM3の統合失調症への関与を機能的に検討するために、mGluR3ノックアウトマウスを作出した。C57BL/6系マウスへの戻し交配後、行動解析を実施した。本疾患のエンドフェノタイブとして知られる運動量の亢進、ワーキングメモリーの障害が見られ、病態への関与が示唆された。
