Research Abstract

NMDA受容体NR2A-2D(ε1-ε4)は長い細胞内C末端を有し、リン酸化によるNMDA受容体チャネルの活性化とともに、scaffoldタンパク、酵素などさまざまなタンパクと会合する。我々は、脊髄での神経因性疼痛の維持にFynキナーゼによるNMDA受容体NR2B(ε2)の1472番目のTyr残基(Y1472)のリン酸化による機能的変化が必要であることを明らかにしてきた。今年度は、このY1472をPhe残基に置換したノックイン(Yl472F-KI)マウス(東京大学医科研・山本雅教授、中澤敬信助教との共同研究)を用いて、神経因性疼痛に関与する機能分子の行動解析、タンパクの発現解析、プロテオミクス解析を行った。野生型マウスの神経因性疼痛では、一次求心性線維の神経終末から遊離されたグルタミン酸がNMDA受容体を活性化させ、脊髄2次ニューロンの細胞内Ca^2+濃度([Ca^<2+>]i)が上昇する。CaMKIIと神経型一酸化窒素合成酵素(nNOS)は共に[Ca^<2+>]iにより活性化され、細胞膜にトランスロケーションする。Y1472のリン酸化が生じないY1472F-KIマウスでも、後根神経節でPACAPの発現が上昇し、nNOSのトランスロケーションによる活性化、ミクログリアの活性化が認められたが、CaMKIIのThr286のリン酸化が障害されていた。このことは、CaMKIIの活性化がこれまで提唱されてきたさまざまな神経因性疼痛の維持機構の最下流にあることを示すものであり、神経因性疼痛の維持は海馬でみられる長期増強など高次脳機能の神経可塑性との相似性が示唆された。
