Research Abstract

γセクレターゼは、アルツハイマー病(AD)発症機構に決定的な役割を果たすAβペプチドの産生の最終段階を担うプロテアーゼである。しかしその単純な阻害はNotchシグナル抑制による副作用を呈することが示されている。これまでにγセクレターゼ活性に対して、『基質特異性』を与える何らかの分子機構の存在が予想されており、APP特異的な切断活性制御機構は副作用のない新たなAD創薬標的分子機構として期待されている。申請者はRNA干渉法(RNAi)の効率がすぐれ、かつγセクレターゼ活性を持つことが当研究室において示されているショウジョウバエS2細胞を用い、ゲノムワイドなRNAiスクリーニング法によりγセクレターゼ活性に基質特異性を付与する分子を同定することを着想・遂行した。本年度は得られた遺伝子群のうちセラミド代謝に影響を与える分子GlcT-1ホモログであるGAMMO4のノックダウンはAβ 42産生を特異的に上昇させた。さらにスフィンゴ脂質およびセラミド代謝経路に関する分子を網羅的RNAiにより解析したところ、やはり複数の遺伝子がRNAiによってG1cT-1と同様にAβ 42産生上昇を引き起こした。またG1cT-1阻害剤であるPDMP処理を行うとやはりAβ 42産生上昇が観察された。これらの結果から、セラミド代謝経路はγ切断特異的に影響を及ぼし、Aβ 42産生量を調節していることが明らかとなった。すなわち、脂質環境、特にセラミド代謝の変化がγセクレターゼ活性に影響を与えAβ 42産生を変化させることが示唆された。
