Research Abstract

DISC1が気分障害の病態とも関係する可能性があり、14-3-3εのSNP-1と気分障害の関連を遺伝統計学的検討と14-3-3ε hetero KOマウス脳を用いモノアミン神経系の免疫組織学的検討を行った。
[方法]
1. 気分障害との関連解析
・ 対象:本研究に関して文書で説明し、同意存取得した双極性障害患者340名、大うつ病患者253名(いずれも診断はDSM-IV TRに則った)と健常者1728名
・ Genotvpe : Taqman法によって行った。
・ 統計学的解析:各グループのHerdy-Weinberg平衡を確認した上で、双極障害、大うっ病性障害と健常者とのAllele頻度ならびにGenotype頻度をx二乗検定で比較した。
2. 14-3-3ε hetero KOマウス脳の免疫組織学的検討
・ KOマウス8匹と対照として野生型マウス6匹を用いた。深麻酔下で灌流固定し脳を摘出後クリオスタットで薄切切片を作成し、抗Tyrosine Hydroxylase(以下TH)抗体を用いてABC法で染色し光学顕微鏡で観察した.。
[結果]
1. Allele頻度において大うつ病性障害のGアレルの頻度が有意差をもって健常者より高かった(p=0.029)。それ以外では有意差を示すものは見られなかった。
2. KOマウス黒質緻密部においてTH陽性神経細胞の密度・面積の減小が認められ、また投射路である前頭前野眼窩面皮質では観察された陽性神経線維の総延長においてKOマウスで有意な減少が観察された。陽性線維の長さに着目すると、20-49μmの比較的短い線維の数が著明に減少し、50-79μmの綿維数も有意な差が認められたが、80μm以上の比較的長い線維では差は認めなかった。
