Research Abstract

これまで、神経原線維変化が形成されるまでにタウはオリゴマー、顆粒状凝集体、線維を形成することを明らかにしている。これらの神経原線維変化が形成される前のタウ凝集形成と脳機能の関係を調べるため前脳領域特異的なalpha CaMKIIプロモーター下で野生型4R2Nヒトタウタンパクを過剰発現するTgマウスを作製し、行動実験・MRI・免疫組織化学の手法によりTgマウスの加齢に伴う脳機能の変化を調べた。その結果、リン酸化タウの蓄積がシナプス消失に関与することが示唆された。タウ凝集体の神経変性への寄与はFTDP17の変異を持つP301Lタウを発現するマウスを解析することで明らかになった。P301L変異を持つタウは野生型に比較して凝集体を形成しやすい。前述した野生型タウTgマウスと同じプロモーターを用いて同じ発現量を示すP301LタウTgマウスを解析した。P301LタウTgマウスはサルコシル不溶性タウを形成していた。神経細胞数はP301LタウTgマウスでnon-Tg,野生型Tgマウスと比べて有意な減少を示した。培養細胞を用いた検討から顆粒状タウ凝集体の形成が神経脱落に関与することが示唆されている。これらの結果はタウのリン酸化、顆粒状凝集体の形成がアルツハイマー病で観られるシナプス消失、神経脱落に関与することを示している。さらに、タウのリン酸化阻害、凝集阻害がアルツハイマー病の進行を阻止する治療法の一つになることを明らかにした。
