Research Abstract

電気細胞融合とは,2種類の細胞の細胞膜を融合させ,これらの細胞の持つ形質を併せ持つ雑種細胞を作り出す手法で,従来法では,2種類の細胞の懸濁液の混合物にパルス電圧を印加することにより行われてきたが,融合の収率が非常に低いことが問題になっていた。そこで,本研究では,細胞径より小さいオリフィス(小孔)を持つ絶縁性のオリフィスシートを挟んで2種類の細胞を対合させ,オリフィスシート両側に置かれた電極にパルス電圧を印加することにより細胞の融合を行う手法を開発した。この手法では,電気力線が絶縁物を通過できずオリフィスに集中するため,電極に印加した電圧のほとんどすべてが2つの細胞の接触点に印加されることになり,細胞の種類・大きさ・配向によらず,非常に高収率(>90%)の融合を実現できる。また,膜電圧が発生するのが細胞の接触点のみであるので,融合は,本質的に必ず1:1となる。
さらに,この手法を用いて,融合後の細胞分裂の経時観察を行ったところ,驚くべきことに,融合細胞が3分裂を行うものが相当の頻度で観察され,そのうち,2度目の分裂においてもふたたび3分裂するものが観察された。このような融合後の細胞の挙動は,ここで開発した高収率融合の手法があればこそ観察可能な現象である。
ここで開発したマイクロ構造を用いた細胞融合の方法は,従来の細胞融合の高収率化のみならず,基礎現象の解明や,細胞核より小さいオリフィスを用いることにより,遺伝情報の混合をともなわずに細胞内の因子のみを他細胞に移植するなど,再生医療やエピジェネティクスにかかわる新しい手法への展開が期待される。
