血小板をヒトES細胞から誘導、高効率産生させる3次元培養技術の確立

Research Abstract

生体内組織幹細胞は高い増殖能、多分化能、自己複製能を兼ね備えた細胞で組織・臓器の発生・修復・維持に重要な役割を担っている。一方、胚性幹(ES)細胞は生体外においてすべての生体構成細胞への分化能力を保持するため、再生医療応用への期待が高まっている。当該年度からは、本特定領域の中での名古屋大学工学部、福田領域代表の研究室との共同研究をさらに発展させてきた。骨髄内部の血管網を模したscaffold deviceを作製することを最終目標として、実際にヒトiPS細胞をソースとした巨核球を用いて、接着性を担保できる培養条件を見いだした。また、材質の改良により、実際の生体内部の循環に伴い発生するshear stress値での血小板放出の定量化をフローサイトメトリーにより行った。現段階では、staticな条件(流れを用いない)での培養から得られる血小板産生数よりも高い産生効率は得られていない為、さらに本研究課題が終了後も、名古屋大学との共同研究を継続する事で、deviceの完成を目指す。さらに、使用する巨核球も成熟度の高まりにより、さらに産生される血小板数も増加する事が期待できる。従って引き続きploidy増加を促進するmediatorの検索・検証も行い、総合的に血小板産生促進のshears tress依存的なdeviceを完成させる。
