Research Abstract

我々はすでに単一巨大リボソーム(GUV)法を用いて、抗菌ペプチドのマガイニン2が脂質膜中にポア(小さな孔)を形成してリポソーム内の物質の膜透過性を増大する現象を研究し、ポア形成の速度定数などの導出に成功している。このポア形成のキネティックパスウエイを解明するために、種々の大きさの蛍光プローブを含む50%ジオレオイルホスファチジルグリセロール(DOPG)/50%ジオレオイルホスファチジルコリン(DOPC)-GUVとマガイニン2の相互作用を研究し、蛍光プローブの漏れの速度の解析からマガイニン2が形成するポアの大きさの評価やポアの大きさの時間変化を研究した。カルセインの場合であれば100%の漏れが起こる濃度のマガイニン2を用いたときに、蛍光プローブのTexas-Red dextran 40kやFITC-BSAの場合は一過性(10秒以下)の急速な少量(全体の10-20%)の漏れしか誘起されなかった。一方、マガイニン2が誘起するTexas-Red dextran 10kやTexas-Red dextran 3kの漏れは2相性を示した。つまり初期の一過性の急速な少量の漏れとそれに続く遅い漏れが観察された。GUV内部の蛍光強度の対数の時間変化の解析から、それぞれの蛍光プローブの漏れに対して、初期状態の漏れの速度定数と最終的な定常状態の漏れの速度定数を求めることができた。これらの結果は、マガイニン2は最初脂質膜に大きなポアを一過的にあけるが、その後ポアの半径は時間とともに小さくなり、最終的には安定な小さな半径のポアに変化することを示している。以上の結果は、マガイニン2の脂質膜中でのポア形成のキネティックパスウエイに関する初めての情報を与える。以上の結果に基づいて、マガイニン2のポア形成の新しいメカニズムを提案した。
