Research Abstract

本研究の目的は,「誘電泳動を利用する細胞操作技術の電気回転現象への応用」により細胞を大量・一括・迅速に制御・回転させ細胞膜表面の受容体の発現量と表現型(フェノタイプ)を解析する手法を確立し,さらに,解析・識別した細胞を分離,捕捉および回収できる簡便なシステムの構築にある.抗CD33抗体を固定化した交互くし型マイクロアレイ電極と平板ガラス基板を厚さ30μmのスペーサを介して挟み込みマイクロ流路構造を有する誘電泳動デバイスを作製した.この流路内に膜表面にCD33抗原が発現している白血球株であるHL-60細胞の懸濁液を導入し,正の誘電泳動により細胞を抗体固定化電極上に配列させた.交流電圧の周波数を変化させ負の誘電泳動を作用させたところ,一部の細胞は電極アレイから電極-電極間に移動したが,ほとんどの細胞がアレイ電極上に捕捉され配列状態を維持した.しかし,CD33抗原を発現していない白血球細胞株であるCCRF-CEM細胞を用いて電極上に配列させたところ,ほとんどの細胞が捕捉されず電極-電極間に移動した.また,電極表面上に固定化する抗体をCD33に特異性を持たない抗マウスIgG抗体を固定化した場合には,HL-60およびCCRF-CEM細胞ともに捕捉されなかった.これは,細胞膜表面上のCD33と電極上に固定化した抗体間で免疫反応が進行したためである.誘電泳動による細胞の配列は約10秒程度で可能であり,トータル3分程度で膜タンパク質を発現している白血球細胞を識別できる.システムを確立するために,細胞の濃度,溶媒の導電率,交流電圧および周波数についても詳細な最滴化を行った.
