Research Abstract

真核生物鞭毛繊毛軸糸のダイニンには内腕ダイニンと外腕ダイニンが存在し、内腕は96nm、外腕は24nmの周期で配列しているが、それらの周期性がどのようにして発生するのかは、まだわかっていない。本研究では外腕の周期的配列の機構を探ることを目的にし、その根元を微小管につなぎとめる複合体(Outer Dynein Arm Docking Complex ; ODA-DC)の構造と微小管との結合性を解析した。これまでの研究では、ODA-DCの3つのサブユニットのうち、微小管との結合に重要なDC1とDC2を培養細胞中で組換え体として共発現させると1:1の複合体を形成し、精製した複合体がin vitroで細胞質微小管上に24nm周期で結合することを明らかにしてきた。今年度は超遠心分析により、DC1とDC2が溶液中でヘテロダイマーを形成することを明らかにした。また、ロータリーシャドウ法による電子顕微法では、ODA-DCが全長約50nmのひも状構造をとることが観察されていたが、この事実と微小管上における24nm周期の結合の関係は説明が困難であった。今回、化学固定した試料を用いたところ、ODA-DCがひも状ではなく、全長20-25nmの、折りたたまれた構造を取ることが観察された。また、微小管と結合したODA-DCの電子線トモグラフィ観察により、24nm周期の結合が確認されるとともに、その結合がこれまで考えられていたような直線状ではなく、分子が微小管の周囲に巻き付いている可能性が示唆された。さらに、軸糸微小管上でODA-DCの結合位置を決定する機能を持つタンパク質の検索を行って、その候補の同定に成功した。これらの発見により、ダイニン外腕が特定の位置で周期的に結合する機構についての知見が大きく前進したと考えられる。
