Research Abstract

Blimp-1と相互作用する因子を検索するためFlagタグのついいたBlimp-1を胚の時期に熱ショジクプロモーター依存的に発現させ、Flag抗体樹脂でBlimp-1タンパク複合体の精製を継続して行い、今回はこの方法で精製した蛋白を質量分析装置で解析した。その結果、ヒストンH4が検出され、機能が明らかになっていないSETドメインなどがヒストンH4に相互作用することによって、Blimp-1の重要な機能の一部が遂行される可能性が示された。
一方、昨年度までに、Blimp-1の発現量を増大させると、蛹化のタイミングが遅くなることを見いだしたので、本年度はBlimp-1遺伝子の欠失変異あるいはPエレメント挿入による変異をヘテロに持つ個体をもちいることで、遺伝子量を半分にする影響を調べた。その結果、Blimp-1遺伝子量が半分になると、蛹化のタイミングが早くなり、Pエセメントを除くことによって回復した。これらの解析結果をから、Blimp-1は蛹化のタイミングを決める分子タイマーの重要な因子で、砂時計の砂のような役割を持つこと考えられた。
また、明らかにしたタイマーシステムが異なる器官で同じタイミングで進行するか調べた。そのために、前蛹期の唾腺、トラキア、中腸、リンググランドでのBlimp-1とFTZ-F1の発現パターンを経時的に蛍光抗体組織染色法で観察した。その結果、両因子ともほぼ同じタイミングで発現することが明らかになったが、器官によっては多少異なる結果が得られた。このことからBlimp-1とFTZ-F1によるタイマーシステムがショウジョウバエ前蛹の様々な器官で基本的に同じタイミングで進行するが、器官によってはその進行速度が異なっており、変態期における器官の違いによる起こる現象の違いに対応している可能性が示された。
