Research Abstract

(1)受容体活性化後の受容体の細胞内移行経路の同定:複数のGPCRを用いてリガンド刺激に伴う受容体の細胞内移行を検討したところ、血小板活性化因子受容体PAFRは速やかに細胞内移行するのに対し、ロイコトリエンB4受容体BLT1は全く細胞内に移行しなかった。様々なキメラ受容体の解析の結果、PAFRの細胞内C末端が細胞内移行に必須のドメインであること、逆にBLT1のC末端はリガンド刺激後の受容体の細胞内移行を抑制することを見いだした。さらに、BLT1のヘリックス8に存在する疎水性アミノ酸LLは、リガンド刺激後のBLT1のC末端領域のリン酸化を抑制していることを見いだした。このLLに変異を加えて、細胞膜にC末端がアンカリングできなくした変異BLT1受容体には過剰なリン酸化が生じる結果、野性型受容体では観察されない受容体の細胞内移行が生じることがわかった。現在、BLT1のC末端に結合し受容体の細胞内移行を抑制する分子、あるいはPAFRのC末端に結合し受容体の細胞内移行を促進する分子の同定を試みている。
(2) 受容体抗体の作成:一般的にGPCRに対する抗体は樹立が困難とされている。本研究ではBalb/c背景の受容体欠損マウスに、受容体を過剰発現した細胞を複数回腹腔内注射し免疫することで、抗受容体抗体価が上昇することを見いだした。これを応用し、抗マウスBLT1単クローン抗体の樹立に成功した。現在、本法を他の受容体にも応用し、抗BLT2抗体、抗PAFR抗体の樹立を行っている。
