Research Abstract

私は、移動神経細胞の過程で、Rho類似G蛋白質の一つであるRac1が重要な役割を果たしていることを明らかにしてきた。そして、その下流でいかなるシグナル伝達機構が関与しているのかを探索し、一つのエフェクターとしてPak1が(1)移動神経細胞に細胞極性を与えること、(2)移動神経細胞が皮質板のもっとも上部で移動を停止させること、に関与していることを明らかにした(Cerebral Cortex, 2009)。また、転写因子Ptflaによって、神経細胞の移動様式が大きく変換されることをこれまでに示してきたが、私はcDNAマイクロアレイの解析から、Ptflaの下流で誘導される様々な遺伝子群を同定した。それらの中には、G蛋白質関連の分子が多く含まれている。特に、Cdc42と細胞の移動方向の決定との関わりあいについても、今後子宮内エレクトロポレーション法などを通して解明して行く。
また、スライス培養系で神経細胞移動を再現し、そこに各種リン酸化酵素の阻害剤を添加する実験によって、神経細胞移動へ関与するリン酸化酵素を多数同定した(JBC2010)。これらのリン酸化酵素のRho類似G蛋白質シグナル伝達系への関与について、現在調べているところである。
