Research Abstract

我々はこれまで、薬物動態に重要な役割を果たす膜輸送体(トランスポータ)に焦点をあて、ヒトにおける薬物トランスポータの発現プロファイルや遺伝子多型と薬物動態との相関について検討を進めてきた。しかし、その発現変動や遺伝子多型では説明できない薬物体内動態の個体差が依然認められることなどから、何らかの制御因子も考慮に入れる必要があるものと考えられた。そこで、ヒト組織における薬物トランスポートソーム構成因子の重要性を明らかにする必要性から本研究を遂行した。昨年度、我々は側底膜有機カチオントランスポータOCT2の発現・機能変動について報告した。本年度は引き続き、腎近位尿細管においてカチオン性薬物の排泄に関わるプロトン有機カチオンアンチポータ(MATE1およびMATE2-K)について研究を進めた。ノックアウトマウスを用いた検討において、カチオン性薬物メトホルミンの尿中排泄速度が顕著に低下すること、この低下がメトホルミンの尿細管分泌の低下に起因することが明らかとなり、MATEがカチオン性薬物の尿細管分泌に主要な役割を果たすことが明らかとなった。また近位尿細管における局在について検討したところ、MATE1及びMATE2-Kともに近位尿細管刷子縁膜に発現すること、さらに側底膜OCT2も同じ上皮細胞の側底膜に発現することが明らかとなった。また、MATEの構造解析をおこなったところ、C末端のアミノ酸残基が膜局在に関与する可能性が示唆され、MATEの膜局在には何らかのタンパク質との相互作用が必要であると考えられた。本研究によって、MATEが薬物腎排泄に重要な役割を果たすこと、またその膜局在にタンパク間相互作用が関与する可能性があることが示唆された。
