Research Abstract

ABCトランスポーターの異常(発現・機能の低下または上昇,異常な細胞内局在など)は種々の疾患の発症や抗癌剤耐性の要因となる.したがって,ABCトランスポーターの細胞内局在やその発現調節の分子基盤を理解することは,細胞の恒常性維持の観点のみならず種々の病態発症機構の理解のためにも重要であるが,ABCトランスポーター分子複合体実体に関しては不明な点も多い.本研究では,疾患関連ABCトランスポーターの膜分子複合体の機能・局在調節機構を解明し,その破綻により発症することで知られる種々の病態との関連性を明らかにすることを目的とし,種々の検討を実施した.その結果,形質膜上に発現することで脂質トランスポーターABCG8は,プロテアソーム依存的に分解されることが明らかになった.また,E3ユビキチンリガーゼの一つであるHRD1遺伝子の導入は,ABCG8単独発現時またはABCG5/8共発現時のどちらの場合においても,脱糖鎖型のimmature ABCG8の発現量増加を引き起こすことが明らかになった.さらに,HRD1欠損変異体を用いた検討から,脱糖鎖型のimmature ABCG8を発現誘導するにはRINGドメインが必要であることが示された.脱糖鎖型のimmature ABCG8は,野生型ABCG5と共存してもヘテロダイマーの形成や形質膜上発現に異常を来すことから,HRD1は,ABCG5/8の形質膜上発現および輸送活性を制御する重要な分子であることが初めて示唆された.
