Research Abstract

GATA2を強く発現する細胞が長期骨髄再建能を有する造血幹細胞であり、骨芽細胞と接触しながら、単独でしかも分裂もせず静かにとどまっていることから、骨髄ニッチによる造血幹細胞の静止・活性化機構の制御に、GATA2が関与していると考えられる。ところが、従来の遺伝子ノックアウトマウス法では、胎児期における重篤な造血器障害にもとづく致死的表現型により、成体骨髄での機能解析は不可能であった。本研究では、発生工学的手法を用いたマウス解析により成獣造血でのGATA2の機能解析を目的としている。本年度は、条件付きGATA2遺伝子ノックアウトマウスを作製し、後天的にGATA2遺伝子を破壊することにより、成獣造血におけるGATA2の機能関与を解析した。その結果、平成20年度で得られたGATA2ノックダウンマウスの結果と異なり、後天的にGATA2を破壊すると、KSL細胞(分化抗原陰性,Sca-1陽性,c-kit陽性細胞)が存在しなくなることを見いだした。これらの結果は、GATA2が全くない状態では造血幹細胞の生存複製が傷害され、その結果幹細胞数が減少すること、一方、GATA2が僅かにでも存在すれば、その生存や複製がある程度保たれること、しかし、GATA2の発現が不足している状態では、幹細胞としての正常な機能ができず、結果的に正常な成熟血球を産生できないことを示唆している。すなわち、造血幹細胞の維持には、複数の分子メカニズムを介したGATA2の機能関与が必要であると考えられる。
