Research Abstract

本研究は精巣の精子形成の元になる幹細胞(精子幹細胞)の移植時におけるニッシェへのホーミング機構に関与する分子メカニズムの解明を目標としている。これまでの研究で研究代表者らはβ1-インテグリンが関与することを同定した。β1-インテグリンは精子幹細胞に発現し、幹細胞ニッシェの基底膜への繋留に関与すると考えられる。一方、ホーミングは基底膜におけるニッシェへの繋留のみでなく、セルトリ細胞への接着やtight junctionの通過など、複数のステップが関与するプロセスであると考えられることから、本年度の研究では他の候補分子(特にrho, rac, cdc42などのG蛋白質シグナルやIntegrin・Cadherinファミリーなど)について調べた。(1) 培養精子幹細胞株(GS細胞)へrho, rac, cdc42変異体の遺伝子導入し、増殖活性や細胞外マトリクス(ラミニン)への接着性を調べたが、特に野生型と差はなかった。移植アッセイによりコロニー形成能についても差は無かった。一方、(2) conditionalノックアウトマウスより精巣細胞を回収し、試験管内でadenovirus (AxCACre)を暴露した後トリプシンで回収し不妊マウスの精巣内へ移植し、コロニー形成能を調べたところ、一部でコロニー形成の低下が見られた。また、(3) ホーミング現象を試験管内で再現するため、セルトリ細胞をフィーダーとした培養系の開発を行った。GS細胞をセルトリ細胞と共培養すると、MEF (mouse embryonic fibroblast)の場合と異なり、フィーダー細胞の下に細胞が潜り込み、コロニー形成することから、これを用いてホーミングアッセイができる可能性が示唆された。
