Research Abstract

私は、移動神経細胞の細胞外シグナルへの反応性が、わずか一転写因子Ptflaの発現によって決定的に変更されるということを示してきた。そこで、Ptflaの下流で発現を誘導される遺伝子が、「神経細胞が細胞外環境に対する応答性」を決めているのであろうと考えて、Ptflaノックアウトマウスで発現が減少する遺伝子、あるいはPtflaの強制発現によって発現が上昇する遺伝子を、cDNAマイクロアレイ解析によって探索してきた。その結果、数十種類の細胞外分泌分子、受容体分子、さらにそれらに関連した細胞内情報伝達分子をコードする遺伝子群を得た。現在、これらの分子が具体的にどのようにして、神経細胞の移動様式に関与しているのかについて、その発現解析や、子宮内エレクトロポレーション法を用いた機能解析によって調べているところである。
また、スライス培養系で神経細胞移動を再現し、そこに各種リン酸化酵素の阻害剤を添加する実験によって、神経細胞移動へ関与するリン酸化酵素を多数同定した(JBC2010)。これらのリン酸化酵素がいかにして細胞内シグナルを受けて神経細胞移動へと寄与しているのか、現在調べているところである。
