ホヤ胚発生における発生運命決定・細胞分化と細胞分裂回数制御の関係

Research Abstract

胚発生において、細胞の発生運命と細胞分裂回数は深く関わっていることが知られているが、本研究では、細胞の発生運命の決定・細胞分化と、細胞分裂停止までの細胞分裂回数との関係を、ホヤ胚を用いて解析する。
1. 発生運命と細胞分裂回数の関係
昨年度は、脊索と神経索に関して解析を行ったが、今年度は、筋肉と間充織に関して解析した。この系ではFGFによる誘導がない場合はデフォルト運命である筋肉に分化し、FGFによる誘導を行うと間充織として発生させることができる。FGFシグナルのありとなしで細胞の運命を変更させたところ、誘導による発生運命決定が同時にその後の細胞分裂回数をも決定していることがわかった。よって、この系でも発生運命と細胞分裂回数が正確にリンクしていることがわかった。
2. キー転写因子の役割
運命決定がなされた直後から組織特異的にキー転写因子群が発現するようになる。これらの因子と発生運命の因果関係はすでに明らかとなっているが、これらの因子が細胞分裂回数を制御しているかどうかを確認した。脊索で発現が開始するBrachyury遺伝子は、脊索分化を司るキー転写因子である。受精卵に合成Brachyury mRNAを注入したあと、64細胞期に目的の割球を胚から単離し部分胚として発生させた。すると、神経索や内胚葉割球も脊索同様に64細胞期以降3回で細胞分裂を停止するようになることがわかった。よって、Brachyury遺伝子は脊索の分化だけではなく細胞分裂回数のコントロールにも機能を果たしていることが示された。
