分裂酵母の細胞質分裂におけるポロキナーゼの役割

Research Abstract

分裂酵母収縮環の形成過程:出芽酵母のアクチン結合タンパク質ABP140のアクチン結合ペプチドLifeactと蛍光タンパク質mRubyの融合タンパク質を分裂酵母内に発現させてアクチンを可視化し、分裂過程でのライブイメージングを行った。分裂期に入ると細胞中央部にアクチンの集積が見られ、そこからアクチンケーブルがダイナミックに伸びたり消えたりするのが見られた。やがて1本のケーブルが細胞赤道部で安定化されて収縮環となっていった。この結果は私達が提唱しているアスター様構造-先導ケーブル仮説を指示するものである。さらに蛍光タンパク質GFPとの融合タンパク質の発現により、分裂に必須なforminであるCdc12がアスター様構造の中央に局在することを見た。
分裂酵母の分裂シグナル関連因子:分裂酵母のキネシンKlp8は分裂期初期に分裂位置に集まる。Blt1というタンパク質はKlp8と共局在する。Klp8とBlt1との関係について詳細に解析を行うため、klp8blt1二重破壊株を作製した。房が破壊株は細胞長が野生型株より長くなる形質を示したのに対し、二重破壊株では細胞長が長くなる形質がやや抑制されることがわかった。Klp8のモーター活性に必須のATP結合部位に変異を入れた変異株においてもblt1破壊株との二重変異による形質の抑制が見られた。Klp8を野生型株で過剰発現すると曲がった細胞が増加するが、blt1破壊株でKlp8を過剰発現すると、さらに多くの細胞が異常形態を示した。以上のことから、Klp8は細胞分裂においてネガティブな機能を持ち、それをBlt1が抑えていることが示唆された。
