Research Abstract

Prader-Willi/Angelman症候群関連遺伝子HERC2のBRCA1に対するユビキチンリガーゼ(E3)活性が細胞周期にどのように関わっているかを解析した。20年度の結果からBRCA1の細胞内半減期はsiRNAによるBARD1の発現抑制によって著明に短縮するが、HERC2の発現を同時に抑制することによって回復し、電離放射線によるDNA損傷後のG2/M checkpoint機能を正常化する結果を得ていたが、HERC2の抑制により、BARD1の発現も軽微であるが回復するため、これがsiRNA効果の減少に起因するものではないことをquantitative real-time PCRにて確認した。複数のsiRNAを用い、全てのsiRNAの実験を3~6回繰り返し、現象が再現されることを確認した。逆にHERC2によるBRCA1のユビキチン化はBARD1によって阻害された。
Crm1阻害剤であるレプトマイシンBにより、HERC2は細胞質から核内に移動し、シャトルタンパク質であることがわかった。原発性乳癌60例のHERC2の発現を解析したところ、HERC2は非癌部乳腺上皮と乳癌の細胞質を主体に発現し、間質には発現していなかった。また、多くの症例でBRCA1の発現とHERC2の発現に逆相関が認められたことから、HERC2はBRCA1の低下が原因で生じる乳癌の発癌にも関連する可能性が示された。これまでの結果は現在投稿中である。
