Research Abstract

動物細胞のMybファミリー転写因子は、c-Myb, A-Myb, B-Mybを含み、細胞増殖と分化に重要な役割を果たしている。c-Mybは、細胞周期のG1/S期の移行に必要な遺伝子や、アポトーシスを防ぐ遺伝子の転写を誘導し、細胞増殖を促進する。一方、B-Mybは紡錘体形成を介して、細胞分裂を制御する機能も有することを、私達は明らかにした。この動物細胞でのMybファミリー転写因子の2つの機能は、細胞増殖と細胞分裂をカップリングさせて進行させる役割を果たしていると、私達は推定している。私達は、Mybを1つしか持たないショウジョウバエでも、Mybが転写制御を介して細胞増殖を正に調節する機能と、紡錘体形成を介して細胞分裂を制御する機能を有するかどうかを検討した。新たに分離したショウジョウバエMyb(dMyb)変異体の1つが、標的遺伝子の転写を誘導する能力は正常であるにも拘らず、幼虫時期に致死となり、細胞分裂装置の異常を示すことを明らかにした。さらに動物細胞のB-Mybと同様に、dMybがクラスリンやフィラミンと結合して、紡錘体形成に関与することを明らかにした。
さらに、B-MybやdMybが、転写制御を介して細胞増殖を正に調節する機能と、紡錘体形成介して細胞分裂を制御する機能とをどのようにして使い分け、カプッリングさせているかを理解するため、B-Mybに結合する因子を探索した。その結果、リン酸化酵素を見出し、このリン酸化酵素が直接B-Mybをリン酸化することを明らかにした。
