Research Abstract

sHspは生物界に普遍的に存在する分子シャペロンで、変性タンパク質の不可逆的凝集を抑制する機能を持つ。通常の温度条件下では分子量12~43kDaのサブユニットが大きなオリゴマーを形成しており、熱によってその構造が可逆的に解離する。解離したsHspは変性タンパク質と結合し、基質を含んだ巨大な可溶性複合体を形成することで、変性タンパク質の凝集を防ぐと考えられている。昨年度に引き続いて好酸好熱性古細菌Sulfolobus tokodaii由来sHsp(StHsp14.0)による変性タンパク質凝集抑制機構の解析を行い、sHspが部分的に解離した状態で変性タンパク質を捕獲し保護するというモデルを構築した。また、分裂酵母由来sHsp(SpHsp16.0)のN末端領域に存在する5つのフェニルアラニンをアラニンに置換した変異体F6A、F7A、F9A、F17A、F20Aを作成し、それらの機能と構造を解析した。各変異体は25℃においては野生型と同様に16merのオリゴマーを形成していることが観察されたが、35℃においてF6A、F7Aの変異体は野生型と異なる溶出時間となり低分子側にピークが移動していた。45℃においては野生型と同様にすべての変異体は低分子側にピークが移動し解離が観察された。これは、SpHsp16.0のN末端領域の先端が、オリゴマー構造形成に関与しており、この部位に変異を入れたことから変異体が解離しやすくなったと示唆される。次に45℃においてモデル基質であるCSを用いた凝集抑制効果を解析した。F17A,F20Aは野生型と同等にタンパク質の凝集を抑制したが、F6A、F7A、F9Aは野生型に比べてタンパク質の凝集抑制能が低下した。この結果は、SpHsp16.0はN末端の特に先端でCSを捕捉し、凝集を抑制していることが示唆している。また、分裂酵母においてsHspと協調して機能すると考えられる種々のシャペロンの発現を行い、特に高効率での発現、精製に成功したHsp90について詳細な機能解析を行った。
