Research Abstract

分解機構の促進という観点からTTRのERAD経路の分子機構解析を行い,小胞体分子シャペロンであるBiPがTTRのERADを負に制御することを見出した.さらに,変異TTRのERADに与えるDerlinファミリー分子の影響を過剰発現系で検討したところ,いずれのDerlinもD18G TTRのERADには影響を与えず,Derlin-1, -2, -3が野生型TTRの細胞外分泌を抑制すること,Derlin-1, -3が野生型TTRおよびD18G TTRの初期合成量を低下させることを見出した.予備検討により,Derlin-1, -3による初期合成量の低下はMG132処理で回復しないこと,TTRのmRNA発現量は各Derlinの発現により変化しないことから,Derlin-1, -3はTTRの翻訳を抑制すると推測される.また,変異TTRの逆行輸送およびユビキチン化に関わる因子の同定を目指し,各種因子のドミナントネガティブ変異体を用いたスクリーニングを行った際に,P97 QQまたはHRD1 C329S共発現時において,本来,非糖鎖付加タンパク質であるTTRがN型糖鎖修飾を受けることを見出した.また,この現象はプロテアソーム阻害剤であるMG132処理においても確認された.さらに,N型糖鎖修飾のコンセンサス配列を検索したところ,TTRのアミノ酸配列中に1つのコンセンサス配列(Asn^<98>-Asp^<99>-Ser^<100>)が存在し,N型糖鎖修飾部位Asn98の変異体(D18G/N98D TTRおよびD18G/N98Q TTR)ではp97 QQまたはHRD1 C329S共発現時に誘導されるN型糖鎖修飾が消失することが明らかとなった.上記の2つの現象は,過去に全く報告がないことから新規性が高く,両機構が細胞内タンパク質の新規秩序維持システムとして寄与している可能性が考えられる.
