Research Abstract

腸管免疫系において炎症反応を司ると考えられる新たなCD4^+ TサブセットであるTh17細胞の存在が明らかになったが、その分化や機能に関しては不明な点が多い。前年度までに、phosphoinositide 3-kinase (PI3K)はTh17細胞分化を正に制御しているが、その分化に重要でありIL-6シグナルの下流に存在するSTAT3には影響しないことを示した。そこで本年度は、それ以外の転写因子に着目して解析を行った。
TGFβシグナルにより発現しTregの分化に重要なFoxp3は、Th17細胞分化でも発現しRORγtの活性を抑制する。しかし、Th17細胞分化においては同時にIL-6シグナルによってFoxp3が抑制されるため、抑制が解除されてRORγtが活性化し、IL-17の転写を促進する。そこでPI3KがTh17分化を正に制御するのは、PI3K活性がFoxp3発現を抑制することが原因であるかについて検討を加えた。まず、野生型CD4^+ TをTh17細胞に分化させる際にPI3K阻害剤を加えたところ、Foxp3発現はむしろ減少した。また、機能的Foxp3を欠損するScurfyマウス由来CD4^+ TをTh17細胞に分化させたが、そこにPI3K阻害剤を加えてもTh17細胞分化は抑制された。さらに、RORγtの遺伝子発現量についても解析したが、PI3K活性を抑制してもRORγt発現は抑制されなかった。以上の結果から、PI3KによるTh17細胞分化制御は、Foxp3やRORγtの発現量調節によるものではないことが示唆された。
