Research Abstract

これまで、我々は、生後マウスの胸腺上皮細胞の一部のみが転写因子Foxn1を発現することを示してきた。本研究では、生後マウスにおける胸腺上皮細胞のFoxn1発現と細胞増殖活性及び機能の関連を、C57BL/6バックグランドのFoxn1::EGFPマウスを用いて出生後のマウス胸腺のEGFP陽性のFoxn1発現細胞について検討した。
胸腺皮質に存在するEGFP陽性細胞のほとんどはLy51陰性であった。髄質に存在するEGFP陽性細胞の多くはAire陽性、UEA-1陽性であった。
また、qPCRによる検討では、UEA1陰性EGFP強陽性(Foxn1発現)細胞は皮質上皮細胞特異的機能分子であるBeta5tとPrss16を選択的に発現していた。さらに、UEA1陽性EGFP強陽性の細胞群は、髄質特異的分子であるAireやRANKを選択的に発現していた。
生後胸腺での増殖性の上皮細胞をKi67の染色で見たところ、髄質に存在するEGFP陽性の細胞群の中にKi67陽性細胞が認められた。BrdU投与による増殖細胞のラベルでも同様の結果が認められた。BrdU投与3週後に認められる細胞回転のきわめて緩やかな細胞群にもEGFP陽性細胞が認められた。
これ等の結果から、Foxn1の発現は、皮質及び髄質の一部の胸腺上皮細胞亜集団の機能維持と上皮前駆細胞の維持に関わっている可能性が示唆された。他方で、胸腺皮質上皮細胞の多くがFoxn1陰性または弱陽性であるがDll4やCCL25を発現して胸腺微小環境の構築に寄与していることもわかった。
