Research Abstract

胸腺細胞の正と負の選択機構では、TCRによる自己抗原認識によって派生するTCRシグナルの差異により細胞の生と死の運命が決定されることから、免疫系のおける自己と非自己認識のモデルとして古くから研究されている。しかしながら、核内でTCRシグナルを感知し、細胞運命決定機構に変換する分子機構は未だ不明である。本研究課題は胸腺細胞のヘルパー/キラーへの系列決定が同様にTCRシグナルにより制御されることに着目し、この系列決定において中心的な役割を果たすThPOK遺伝子発現制御機構を解明することで、TCRシグナル下流で細胞運命決定を制御する分子機構の解明を目的とするものである。本年度は、ヘルパー系列特異的なThPOK遺伝子の発現に必須であるThPOKサイレンサーの機能解析を行い、ThPOKの発現誘導にはMHCクラスII拘束性のTCRシグナルによるThPOKサイレンサーの機能解除が必須であり、ThPOKサイレンサーがTCRシグナルの標的DNA配列であることを支持する結果を得ることが出来た。次に、生化学的手法にて、ThPOKサイレンサー結合因子の精製を試み、数種類の候補分子を得た。次に、ChIP法等の分子生物学的手法により、少なくとも4種類の因子は真の結合因子であることが確認出来た。更に、これら分子のThPOK遺伝子発現における機能を解析する目的に、発生工学的手法を用い遺伝子改変マウスの作製に着手した。このような胸腺細胞の分化運命決定に関与する核内分子の同定は、今後自己と非自己の識別機構の解明に繋がる研究成果といえ、その一部を国内、国際学会にて発表した。
