Research Abstract

適応免疫応答の中枢を担うT細胞は胸腺の中で増殖・分化するが、この際ランダムな遺伝子再編成により作り出された多様なT細胞の中から、有用な細胞のみを生存させる"正の選択(ポジティブセレクション)"と、自分の身体の成分と強く反応する細胞を消去する"負の選択(ネガティブセレクション)"の2段階の選択が行われる。これまで、胸腺髄質上皮細胞(mTEC)において組織特異的抗原を発現させる転写因子AIREの発見やmTECの分化に必要なシグナルの同定など負の選択の機構の理解は進んでいたが、胸腺皮質上皮細胞(cTEC)で行われる正の選択の具体的なメカニズムは殆ど不明であった。
申請者らはcTECに特異的に発現するプロテアソームの新規触媒サブユニットβ5tおよび特異な酵素学的性質を持つ"胸腺プロテアソーム"を発見し、CD8^+ T細胞の分化に必須であることを見出した。プロテアソームによるタンパク質分解産物は主要組織適合性複合体(MHC)クラスIに提示される抗原ペプチドとして利用され、CD8^+細胞傷害性T細胞が自己と非自己を識別するための旗印となることから、胸腺プロテアソームが正の選択のための特別なペプチドレパートリーを作り出している可能性が強く示唆される。本年度はβ5tが胸腺特異的に発現する機構に関して解析を行った。まず、ルシフェラーゼアッセイを用いたプロモーター解析を行い胸腺特異的な発現に必要なプロモーター領域を同定した。次にβ5tを発現していない細胞でβ5tを発現させるような転写因子を探索し、候補蛋白質としてFoxN1を同定した。現在、cTEC細胞において実際にFoxN1がβ5tの発現に関与しているのかどうか検討中である。
