Research Abstract

C型肝炎ウイルス(HCV)は感染後、高率に持続感染化し、慢性肝炎、更に高率に肝細胞癌を発症する。HCV感染ヒト肝臓組織ではHCVが持続的に複製し、特異的細胞障害性T細胞(CTL)が誘導されているにも関わらず、ウイルス感染細胞が完全には排除されないことが知られている。このことから持続感染が成立する理由の一つとしてHCVに対して宿主側が免疫寛容状態になっていることが推測される。我々はCre/loxPシステムでHCV遺伝子を導入したトランスジェニック(Cre/loxP/HCV-Tg)マウスと、IFN誘導性のCreを発現するトランスジェニック(Mx-Cre-Tg)マウスを交配させることにより、任意の時期にHCV遺伝子をスイッチング発現するトランスジェニック(Cre/loxP/HCV-MxCreTg)マウスを作製した。Poly(I:C)投与により発現させた肝臓内のHCV蛋白は、完全に排除されることなく、600日以上の持続的な発現が確認された。形態学的には遺伝子発現後6~7日で壊死を伴う比較的高度の炎症細胞浸潤が認められ、その後、門脈域を中心とした弱い炎症反応が持続的にみられた。他の病態としては肝細胞の膨化、脂肪変性、グリコーゲン変性、肝細胞索の乱れ、線維化、肝細胞癌およびリンパ腫などがみられた。脂肪変性1に関しては遺伝子発現後21日から認められた。以上より、スイッチング発現システムを樹立したことにより、発生段階でのHCV遺伝子の発現はなくなり、胎仔に影響を与えることなくHCV持続感染時に似た免疫反応状態をつくることができた。
