Research Abstract

真核細胞内ではmRNAはつねに特定の蛋白質との複合体mRNPとなっている。そのため、mRNP構成因子はmRNAの安定性や翻訳の調節に重要な役割を果たす。免疫染色やGFP-融合蛋白質の観察により、mRNP構成因子の多くが、細胞質で顆粒状に局在することがわかっている。本研究では、これら細胞質mRNP顆粒(P-bodyやストレス顆粒)の形成と分解の機構について調べた。P-bodyとストレス顆粒では、共通に見られる構成因子もあるが、一部、各顆粒特異的な構成因子もあることが知られている。1)カエル卵母細胞の母性mRNPに特異的な構成因子P100/Patlaは翻訳を抑制する活性を持つが、その局在を見ると、もう一つのmRNP構成因子RAP55Aとは異なって卵母細胞で顆粒状の局在は見られなかった。体細胞に発現するパラログであるPatlbはP-bodyに局在した。
2)ヒトRAP55Aは、内在性蛋白質の免疫染色でP-bodyおよびストレス顆粒に局在が見られるが、HeLa細胞に高発現させると大きな顆粒の形成がみられた。これらの顆粒には、P-bodyおよびストレス顆粒に特異的なマーカーが両方とも存在していた。3)RAP55AのパラログであるRAP55Bも、HeLa細胞に発現させるとP-bodyに局在し、高発現で大きな顆粒の形成がみられることがわかった。一方、RAP55AをノックダウンするとP-bodyが消失するので、これらの結果は、細胞内でのRAP55Aの量がmRNP顆粒形成の調節に重要であることを示す。
