Research Abstract

本研究の最終的な目的は、遺伝学的手法と構造生物学を組み合わせて(=構造遺伝学)、真核生物の繊毛・鞭毛の中で多数のダイニンが協調して運動をする仕組みを理解することである。
ダイニン分子の構造的な標識はATPにビオチン標識を入れることで、頭部の回転が少ないことが観察されつつある。またダイニンの微小管結合部位であるストークと微小管の複合体のクライオ電子顕微鏡による高解像度解析が進み10オングストロームを超える構造が得られた。さらに、鞭毛の中心微小管についても三次元再構成の方法が確立しつつある。
また、鞭毛による細胞駆動を評価するための三次元トラッキング顕微鏡の技術を確立した。現在最大2分間、一つのクラミドモナス細胞を追いかけながら鞭毛の動きを観察できる。その動きについてコンピュータによる速度、周期、鞭毛の長さ、細胞の大きさなどの多変量解析が進行中で、論文を執筆中である。これによって、クラミドモナスの2本の鞭毛が同調したり非同調したりする仕組みを解明する事が可能になるだろう。
最後に、本研究によって得られた成果を元に、研究代表者・吉川は最先端・次世代研究開発支援プログラム「新しいイメージング手法による鞭毛の分子機構」の受給を受ける事になった。従って、この基盤研究は、H23年度よりその内容を発展させて最先端・次世代研究開発支援プログラムに引き継がれることとなった。
