プレT細胞受容体を介するT細胞分化機構の解析

Research Abstract

・プレTCRα鎖(pTα)ノックインマウスの樹立
マウスpTαゲノムを単離し、荷電アミノ酸(D22,R24,R102,R117)をそれぞれアラニン(A)に変異させ、intron部分にloxP-neo-loxP、長腕の外側にDTカセットを挿入したベクターをES細胞(Bruce4)に導入し、相同組み換えクローンを取得した。ES由来のキメラマウス7匹をC57BL/6に交配し、germline transmissionが完了した個体を樹立することができた。更に、当該マウスを交配してKI/KIマウスを樹立し、プレTCRを介するT細胞分化シグナルが障害されていることを見出した。T細胞分化に必須のレセプター、プレTCRが、リガンド非依存的にユニークな自己会合能を介してシグナルを伝えていることを個体レベルで示した初めての例となった。
・プレTCR特異的なシグナル伝達経路の解析
TCR下流でNFkB経路の活性化に必須の役割なアダプター、Bcl10欠損マウス、またNFkB2経路に必須なNIKの変異型、NIK^<aly/aly>マウス、及び二重変異マウスにおいても、プレTCRシグナル、プレTCRによる胸腺細胞分化は全く正常に認められることが明らかとなった。また、βセレクションに伴って誘導される遺伝子群に関しても、これら変異マウスにおいて全く正常に誘導されることが判明した。この結果より、成熟TCRにおいては必須の役割を果たすNFkB経路が、プレTCRにおいては必要でない、すなわち、未熟TCRと成熟TCRとでは、細胞内の伝達の機構が全く異なることが判明した。構造的には類似した受容体を介するシグナルが、分化段階によって劇的に異なる応答の惹起へと繋がる分子機構を解く手がかりとなると考えられる。
