Research Abstract

ほ乳類の細胞のテトラヒドロビオプテリン(BH4)産生には二つの経路が存在することが知られている:GTPCHによって始まるde novo synthetic pathwayと存在するダイハイドロビオプテリン(BH2)を利用するsalvage pathwayである。BH4のde novo合成の律速酵素GTP cyclohydrolase I(GTPCH)は血管平滑筋においてはlipopolysaccharide(LPS)によって誘導型一酸化窒素合成酵素(iNOS)と共誘導され、このことがBH4産生そしてNO産生に必須である。外因性BH4の供給にはBH2がdihydrofolate reductaseを経てBH4となるsalvage pathwayが主な役割を果たしていると考えられた。外因性のBH4としての効率はBH4およびそのprecursorとしてのBH2/sepiapterinを用いて比較検討したが、その効率はsepiapterin>BH2>>BH4の順で、細胞膜の透過の機構がどうやら重要な問題であると考えられた。今年度は、以上のことを、補足し確認する実験を行ってきた。
