Research Abstract

C57BL6マウスcDNA由来のtruncated TGFβ1遺伝子を導入したNIH3T3細胞を培養し、truncated TGFβ1タンパクを含む培養上清を得た。これを、各種口腔癌細胞株(NA,CA9-22,HSC-4など)の培地に添加してinvasion assayを行ったが、それらの浸潤能に影響はなかった。
C57BL6マウスより骨髄を採取し、これをIL-4とGM-CSF存在下で14日間培養し、培養樹状細胞(cultured DCs)とした。ここに、マウス肺癌細胞株3LLのlysateを添加してさらに5日間培養しlysate-pulsed DCsを得た。C57BL6マウスの3LL移植腫瘍モデルを構築し、形成された背部腫瘍の周囲にこれらのlysate-pulsed DCsを2x10^5/tumorで局所注射し、3週間経過観察したところ、局所注射なしの腫瘍と比較し、その増殖は著明に抑制された。腫瘍の病理組織学的検索においても、lysate-pulsed DCs注射群の腫瘍内には著明なT細胞の浸潤が認められ、かつ腫瘍細胞の大部分はapoptotic deathの状態であった。現在、このモデルにおけるtruncated TGFβ1/TGFRの影響を評価している。
