Research Abstract

本研究の目的は,特定の神経細胞の膜動態を得る為に,実際の細胞膜上にあるイオンチャネルや受容体の数理モデルを計算機上に構築し,細胞膜動態の特性を理解することにある.そして,ダイナミッククランプ法という手法を用いて,細胞内に電流を注入することで,架空のイオンチャネルを細胞膜上に,ノックイン,および,ノックアウトする技術を発展させた.この技術を用いて,細胞膜上に特定の細胞膜動態を作成することを興奮膜におけるリーバース・エンジニアリング(RE)法とよぶ.その為に,本研究では,in silicoにおける計算機によるシミュレーションとwetな実験系によるモデル検証を行なった.そして,どの様な生化学反応とイオンチャネルが本質的に膜動態の形成に最も重要であるかを明らかにしようとした.特に,本研究課題では,ドーパミン作動性細胞の膜動態を得るためのRE法の開発を行った.さらに,実験の効率を大幅に向上させるためにon-chip集積型デバイスの開発を行う.特に,従来の細胞のパッチクランプ法では,一回の実験において,少数(2~4)の記録用ガラス電極のみしか計測に用いることができないために,細胞集団の計測には不向きであった.そこで,本研究で開発を目指したのは,ガラス電極の先端をアレイ状に配置した平面デバイスを,微細加工技術を用いて製作することであった.そして,その細胞インターフェースとなる先端部に通じる後細流路を製作して,アレイ型パッチクランプ法の原型となるデバイスを製作した
