シナプス前終末内ミトコンドリアによるシナプス可塑性の制御機構

Research Abstract

1.海馬スライス標本のCA1領域において、細胞外電位記録法を用いて興奮性シナプス応答を記録した。NMDA受容体を阻害した状態で5Hz、3分の刺激を与えると、刺激終了後に二相性の短期可塑性が誘導されることを見出した。早い相では2発刺激促通の減少を伴う短期増強が消失していくところが観察され、遅い相では短期抑圧が徐々に回復していくところが観察された。ミトコンドリアの機能を変化させる種々の薬物によりこの短期可塑性が修飾されることから、これらの短期抑圧にミトコンドリアのカルシウム放出能やエネルギー産生が関与していることが明らかとなった。これまで知られていなかった神経機能におけるミトコンドリアの新たな役割を解明することができた。シナプス前終末のミトコンドリアの機能を阻害した遺伝子改変マウスなどを用いて、さらに解析を進めている。
2.海馬歯状回においては、シナプスが1Hz程度の比較的低頻度で活性化しても、顕著なシナプス抑圧が起こることが知られているが、その抑圧発現機構についてはよくわかっていなかった。海馬歯状回スライス標本において、電気生理学的手法を用いて、シナプス伝達に関する種々のパラメーターを検討したところ、このシナプス伝達抑圧には、シナプス後細胞での変化は関与しておらず、シナプス前性の修飾により発現していることがわかった。さらに、シナプスの種類により神経伝達物質の放出確率の低下とシナプス小胞プールの枯渇のいずれかがおもな原因であることが明らかとなった。
