Research Abstract

酸処理によって金属触媒を除去した多層カーボンナノチューブ(MWCNT)をウシ胎児血清(FBS)を用いて水中に分散した(FBS-CNT)。FBS-CNT非含有培地中の場合(Control)と比較して、培地中のFBS-CNT濃度が1.0mg/mlの場合では増殖が抑制された。一方、0.1mg/mlの場合ではほぼ同程度の増殖を示したものの、形態においては長く伸びた仮足および細胞上に伸びた仮足が観察され、顕著な差異が認められた。蛍光試薬(FITC)をラベル化したBSA分散MWCNT(FITC-BSA-CNT)含有培地中で培養した細胞を蛍光顕微鏡で観察したところ、FITCの緑色発光が観察され、細胞内に取り込まれたFITC-BSA-CNTが認められた。FITC-BSA-CNTは細胞質および核の両方に局在し、特に核とその周辺に局在しやすいことがわかった。また、導入率(FITC-BSA-CNTが取り込まれた細胞数/全細胞数×100)は、曝露12時間の場合が最も高い6%を示し、その後は減少した。これは、CNTは細胞内に存在するものの、FITCの発光が衰退したためだと考えられた。同様に、FITC-BSA-CNT含有培地中で培養した細胞に電位印加を行い細胞膜障害を評価したところ、Controlと比較して、FITC-BSA-CNT曝露細胞の場合では全ての曝露時間において印加時間が約2~7分短くなることが示された。このことから、蛍光顕微鏡による観察では解らなかった曝露12時間以降の細胞内CNTの存在が明らかとなった。以上のことから、MWCNT分散培地中で培養した場合、MWCNTは細胞内に取り込まれ、細胞形態・増殖に影響を及ぼすことが明らかとなった。また、MWCNTが取り込まれた細胞を電位印加によって高感度に検出できることが示された。
