Research Abstract

同じ植物種でも、育つ環境で植物体は大きくも小さくもなる。植物のサイズは栄養条件や温度、光、水といった要素だけに左右される訳ではなく、どのような植物が周りに植えられているのか,あるいはどのような密度で植えられているかによっても異なる。シロイヌナズナのある遺伝子(PCaP)の破壊株を得て、その種子を寒天培地の上に様々な個体密度で播いたところ、低密度播種の場合は野生株と変化がないのに、高密度の場合は野生株に比べて遺伝子破壊株は生育が抑制され小さな個体となった。そこで、このPCaP1およびPCaP2遺伝子産物がどのような分子であるかを解析し、次の知見を得た。(1)PCaP1とPCaP2はリジンやグルタミン酸に富む酸性タンパク質であり、膜貫通領域をもたない。しかし、N端にミリストイル基が共有結合することで細胞膜に安定に結合する。(2)カルシウム、ボスファチジルイノシトールリン酸(PtdInsP)、カルモジュリン/Ca複合体との結合能を有し、カルモジュリン/Ca複合体の結合は、PtdInsPとPCaPとの結合を解除し、結果としてPtdInsPが遊離する。(3)PCaP1は根、シュートのいずれの組織でも発現し、とくに表皮細胞でのタンパク質蓄積量が多い。(4)PCaP1遺伝子の欠失株では、ある情報伝達分子関連遺伝子の発現が増える。これらの知見を総合して、接触刺激を含む外部情報が細胞質カルシウム濃度変化となり、その変化が、細胞膜のPCaP1を介してPtdInsPへの情報へと流れていくという作動モデルをまとめた。
