Research Abstract

Runx2は、骨芽細胞・軟骨細胞分化に重要な役割を果たしている。そして、Runx2の時間的・空間的発現および活性制御が、骨芽細胞・軟骨細胞分化および骨格形成・維持プログラムに必須である。EGFPとDsRedによりRunx2の発現と活性を同時に観察できるトランスジェニックマウスを作製し、その中足骨を用いた器官培養により、Runx2の発現と活性を調節できる化合物のスクリーニング系を確立することを目的とした。本年度は、異なるRunx2プロモーター領域を含むRunx2プロモーター・EGFPトランスジェニックマウスを7系統樹立した。それぞれの系統でのEGFP発現を、GFP抗体を用いた免疫組織染色で調べた。胎生16.5日、1週齢、4週齢で、骨芽細胞、軟骨細胞おける発現パターンを検討、骨芽細胞、軟骨細胞におけるRunx2発現誘導を検討するのに適した系統を決めた。これらの系統の中足骨器官培養を用いて、FGF2、FGF18、BMP-2、nogginの効果を検討した。GFP強度でそれぞれの因子の骨芽細胞、軟骨細胞におけるRunx2発現誘導能を明らかにすることができた。また、これは、GFP抗体を用いた免疫組織染色法で確認することができた。一方、Runx2の結合配列をタンデムに6つ並べたDNAにminimal promoterをつないだDsRedトランスジェニックマウスを作製したが、DsRedの高発現マウスを得ることはできなかった。今回作製した、Runx2プロモーター・EGFPトランスジェニックマウスは、Runx2の発現を調節できる化合物のスクリーニング系に有用であり、骨粗鬆症や、変形性関節症治療薬の開発に大きく貢献すると考える。
