Research Abstract

細胞移植による変性聴神経の置換再生の可能性を前年度に引き続いて検討した。具体的には、圧迫変性後の聴神経にN33を移植する実験を前年度に引き続いて実施した。本年度は、聴神経変性と共に生じたgliosis内に細胞を移植する実験を実施した。Gliosis部分には、astrocyteなどによって産生されるchondroitin sulfate proteoglycansが豊富に発現していることを証明した後に、これを消化する酵素であるchondroitinase ABCを移植細胞と同時に投与する実験を行った。その結果、細胞生着と分化が促進されて移植細胞が双極性を取って末梢方向に遊走する像をとらえることができた(論文準備中)。
また聴神経内に生じるgliosisが聴神経鞘腫摘出術後に遅発性に生じてくる難聴の原因である可能性を論じた論文を投稿し、Mechanical stress-induced reactive gliosis in the auditory nerve and cochlear nucleus, Journal of Neurosurgeryと題して公表した(DOI: 10.3171/2010.2. JNS091817)。
この研究成果とは別に、さらに前年度までの成果として聴神経圧迫変性後に蝸牛神経核(cochlear nucleus)に生じる変化を「定量的に」評価して投稿をした論文が公表された(Sekiya, T, Canlon, B, Viberg, A, Matsumoto, M, Kojima, K, Ono, K, Yoshida, A, Kikkawa, YS, Nakagawa, T, Ito, J.
