Research Abstract

研究計画調書の内容に従って研究を遂行した。すなわち、転写振動を支えるエピジェネティックな時刻管理法則の同定と生体リズム調整剤の開発に向け、時計蛋白質PER2を含む転写調節複合体の精製、および脳内の中枢時計である視交叉上核(SCN)において発現する機能未定のG蛋白質共役型受容体群およびそのシグナル調節因子の網羅的検索と機能解析を行った。そのなかで早朝に特に強くSCNに発現し、G蛋白質を介した細胞内のシグナル伝達制御に関与する因子に着目し、その機能解析を遺伝子改変マウスを用いて進めた結果、本因子を介した時刻依存的なサイクリックAMPシグナルによって時計遺伝子の発現リズムが調節されることを見出した。また、時計遺伝子Per1プロモーター発光遺伝子トランスジェニック動物と極微弱光測定技術を応用したSCNスライス培養下における連続リズム測定系を用いてSCNに作用する化合物を検索する過程において新たにAMPA型グルタミン酸受容体を介したシグナルがSCNの時計の時刻をシフトさせることを明らかにした(Mizoro et al, Plos One, 2010)。またさらに時計遺伝子Cry欠損によって生体リズムに異常を来たしたマウスが食塩感受性高血圧を呈することを見出し、その病因が分子時計に制御される新規のアルドステロン合成系酵素Hsd3b6の過剰発現にあることを突き止めた(Doi et al, Nature Med., 2010)。本酵素はヒトにも保存されており、これまで原因不明の難治性疾患として知られていた特発性高アルドステロン症の原因となる可能性がある。疫学調査から高血圧の罹患率は昼夜交代勤務者において高いことが知られていたが、実際に生体時計と高血圧を結びつける分子機序は不明であったため、これらの研究成果は、疾患における生物時計の意義付けを大きく変える重要な知見を提供したといえる。
