Research Abstract

申請者はこれまで、腫瘍血管新生因子VEGF遺伝子のエクソン1領域に、蛋白質をコードしないRNA(以下VEGFノンコーディングRNA)が内蔵されていること、このVEGFノンコーディングRNAが「癌促進性RNA」として機能することを明らかにしてきた。本研究では、VEGFノンコーディングRNAによる腫瘍抑制遺伝子群のサイレンシング機構を解明することを目的とし、以下の研究成果を得た。
(1)VEGFノンコーディングRNA制御系とVEGF蛋白質シグナルからなるネットワークの解明
VEGF標的治療薬(モノクローナル中和抗体、VEGF受容体阻害剤)によってVEGFノンコーディングRNAが細胞レベルおよびマウス個体レベルで誘導され、VEGF標的治療薬に対する抵抗性が獲得されることを明らかにした。また、この誘導は、VEGF遺伝子の転写からノンコーディングRNAの転写へスイッチングすることによっておこることを見出した。
(2)VEGFノンコーディングRNAによる癌幹細胞化
In situ hybridyzation解析の結果、VEGFノンコーディングRNAはヒト大腸組織の幹細胞に強く発現しており、アデノーマ組織及びカルシノーマ組織ではより強く発現していることを見出した。また、ヒト大腸癌細胞株にVEGFノンコーディングRNAを安定発現させると、マウス移植モデルにおいて悪性腫瘍を形成することを確認した。
