We report that Arg-Gly-Gly domain (RGG) in EWS binds G-rich single strand DNA fold in G-quadruplex DNA while it cannot bind G-rich double and single stranded DNA. Inhibition of DNA polymerase on a template containing G-quadruplex in the presence of RGG occurred in an RGG concentration-manner by forming a stabilized G-quadruplex-RGG complex. Moreover,」 specific binding of RGG to G-quadruplex DNA is dictated by hydrogen bonding of arginine in RGG.
