Outline of Research at the Start

Developing new methods of mosquito control is essential, but requires improving current knowledge of basic mosquito biology. This project focuses on exploring the mechanisms underlying mosquito hearing behaviours, which form a key part of courtship, in order to develop novel control methodologies. We will investigate how the circadian clock influences mosquito hearing capabilities, and the effect of clock gene knockout on mating. After identifying genes which affect mating behaviour, we will test whether exposing mosquitoes to insecticides which interfere with the clock can inhibit courtship.

Outline of Annual Research Achievements

蚊は吸血することで感染症を媒介し、多くの人を死に至らしめる。蚊の吸血は、交尾後のメスが産卵のための栄養源を得る行動である。よって、蚊の被害を防ぐ方策の一つとして、配偶行動への介入が考えられる。本研究では、世界各地でデング熱やジカ熱を媒介するヤブ蚊類2種(ネッタイシマカ、ヒトスジシマカ)を対象に、聴覚を介した配偶行動の原理とその神経基盤を理解し、繁殖制御への新たな道を探ることを目的とする。
前年度までに、研究室内でオスの聴覚を介した配偶行動を解析できる実験系を立ち上げた。そこで当該年度では、様々な温度域において音源への接近飛行行動を測定した。18度から28度までの温度域で行動を解析できる実験系を整備して順次測定を行い、温度と接近飛行行動との関係性を見出すことに成功した。
上記の解析結果から、聴覚器の感度が、温度変化に対応している可能性が提案された。そこで、蚊の聴覚器の振動を計測することが可能なレーザードップラー振動計を用いて、様々な温度域における聴覚器振動の特性を解析した。その結果、温度変化による接近飛行行動の変化と良い相関を示す、聴覚器振動の温度に依存した特性変化を検出した。
さらに、脳から聴覚器への遠心性神経投射による制御の可能性を追求した。オスとメスの聴覚器からRNAを抽出し、遠心性神経投射に関わる可能性がある神経修飾物質の受容体の、聴覚器での発現を解析した。その結果、複数の受容体遺伝子が聴覚器で発現することを見出した。

Reason

蚊は配偶行動時に羽音を用いた求愛交信を行う。そのため、彼らの配偶行動の成立には、精緻な聴覚システムが必須である。本研究の目的は、ネッタイシマカとヒトスジシマカ、という2種類のヤブ蚊を対象として、配偶行動に重要な機能を持つ聴覚系の動作原理や聴覚行動の神経基盤の理解を進めることで、繁殖制御への新たな道を探ることである。
当該年度では、前年度に立ち上げた、聴覚を介した配偶行動の解析システムを用いた解析を実施した。様々な温度域において、羽音を模した音源への接近飛行行動を測定した。その結果、温度変化により、接近飛行行動を誘発する音の周波数が変かすることを見出した。
また、蚊の聴覚器の振動を計測する実験系を確立し、温度変化による接近飛行行動の変化と良い相関を示す、聴覚器振動の温度に依存した特性変化を見出した。
さらに、脳から聴覚器への遠心性神経投射による制御の可能性を追求した。遠心性神経投射に関わる可能性がある神経修飾物質の受容体群の、聴覚器での発現をRT-qPCR法により解析した。その結果、複数の受容体遺伝子が聴覚器で発現することを見出した。
以上、当該年度の研究は、昆虫の中でも特に発達した蚊の聴覚機能の新たな特性の発見につながった。

Strategy for Future Research Activity

今後は、聴覚器での発現を見出した、神経修飾物質の受容体群の遺伝子発現を抑制した個体を作成し、聴覚システムへどのような影響が出るかを解析する。聴覚を介した配偶行動や、聴覚器の振動特性への影響を調べる。また、温度受容を担う分子実体の同定も進める。ショウジョウバエで同定されている一連の温度受容体を候補として、遺伝子発現を抑制した個体を作成し、行動の温度依存性への影響を解析する。
