Outline of Research at the Start

高速AFMの誕生により、生体分子の形状とダイナミクスの同時観察が初めて可能となった。しかし、現在の高速AFMの時間分解能では観察できない生命現象の諸過程はまだまだ多い。そこで本研究では、現在の高速AFMの時間分解能を律するデバイス群を大幅に改良し、より高い時間分解能を持った次世代型高速AFMの研究開発を推進する。また、開発した新規の高速AFMを用いてバイオ応用研究を進め、精製した生体分子および、オルガネラや真核細胞表面の生体分子の詳細動態解析を通じて、開発した顕微鏡の性能を示すとともに、生物学的に意義深い新知見を得ることを目的とする。

Outline of Annual Research Achievements

本年は、昨年度までに開発に成功していた高速振幅計測器を実用化するために、広帯域の電子回路の製作得意とする企業とともに更なる広帯域化と操作性を向上させた電子回路を共同開発した。自作したものの帯域の数倍の高速化が見込まれている。また、昨年度までに開発していた超小型高速カンチレバーの変位をより精密に計測するために、変位計測のためのレーザースポットの微小化に取り組んだ。レーザースポット径を精密に計測する手法を開発し、レーザーダイオード、コリメーションレンズ、対物レンズを検討することで、従来のスポット径の21%程度にまでスポット径を微小化することができた。さらに、昨年度までに考案していた逆伝達関数法とアクティブダンピング法を組み合わせた電子回路（共振コントローラー）では、Zスキャナーの共振周波数とQ値を一緒にコントロールできることが実証できたため特許出願を行った（特願2022-141581）。
また、開発途中の次世代型高速AFMを用いて、バイオ応用研究を進めた。たとえば、慢性腎臓病や骨粗鬆症などに関与するビタミン D 代謝酵素の過剰発現を抑えるために、ビタミン D 代謝酵素とDNAアプタマーの複合体の観察に成功した（ACS applied materials & interfaces誌）。また、新規のゲノム編集タンパク質CRISPR-Cas3が、狙ったDNA配列周辺のDNAを大規模に切断する様子の直接観察に成功した（Nat. Commun.誌）。その他にも、微小管結合タンパク質であるEB1とSka1の複合体（J. Biol. Chem.誌）、アクチン結合タンパク質であるRng2がミオシンIIの運動の下げること（Life Science Alliance誌）などを明らかにした。これにより開発した顕微鏡装置の性能を示すとともに、生物学的に意義深い新知見を得ることができた。

Current Status of Research Progress

Reason

現在の高速AFMの時間分解能を律するデバイス群を大幅に改良し、次世代型高速AFMの研究開発することを推進している。開発すべき要素技術は、①Zスキャナー、②デュアルZスキャナーシステム、③カンチレバーの振動振幅計測回路、④超微小カンチレバー、⑤超微小カンチレバー用の変位検出系である。これらを全て高速化・高S/N化することで、装置のZ方向の力制御に関わる周波数帯域（fB）を現行の装置の約7倍である約500 kHzにまで高めることを目標としていた。①、③、④、⑤に関する開発については、目標性能を達成することができ、①については、電子回路を用いた共振コントローラーの開発にも成功したことで、さらなる高速化が期待される状況である（特願2022-141581）。③については、企業と共同してより高い性能と操作性を持った計測器の開発を進めている。④は微小カンチレバーの変位をより高精度に計測するために、変位計測用のレーザースポットを従来の21%程度までに微小化することに成功した。②は、回路シミュレーターを用いたシミュレーション実験を行い、原理的に実現可能であることが確認できているが、本年度は進めることができなかった。一方で、真核細胞の高速AFM観察の系の立ち上げに成功した。また、開発途中の次世代型高速AFMを用いて、いくつかのタンパク質系でバイオ応用研究も進めている。たとえば、細胞内での機能が良く分かっていないSMCタンパク質について、そのサブ分子形状まで識別しながらDNAと結合する様子を可視化することに成功している。ヌクレオチド条件で結合様式が変化することも見えているため、その分子メカニズムに深く迫れることが期待される。

Strategy for Future Research Activity

高速AFM装置の走査速度を大幅に改良するために、昨年度までに本研究では、1.1 MHz程度の共振周波数を持つ超高速Zスキャナー、共振周波数と共振のQ値をコントロールできるZスキャナー共振コントローラー、カンチレバーの振動振幅を振動周期の5倍程度の速度で計測できる高速振幅計測器、液中で10 MHz程度の共振周波数をもつ超小型カンチレバー、超微小カンチレバーの変位を高精度で検出する光学システム、超微小カンチレバーのための加振システムを開発してきた。本年は、まだ改良の余地がある加振システムをさらに改良することで、超微小カンチレバーを10 MHz程度で安定に加振できるようにする。光学システムでは、カンチレバーの背面に照射するレーザースポット径の面積を従来の21％程度にまで縮小することに成功しているが、レーザー素子や対物レンズの組み合わせを検討することでさらなる縮小化を目指す。また、上記のデバイスを全て搭載した次世代型高速AFMをくみ上げ、フィードバック制御帯域や観察対象への低侵襲性などといった性能の評価を行う。
また、これまで開発してきた最速の高速AFM装置を用いて、バイオ応用研究に取り組む。細胞骨格（アクチン線維や微小管）に結合するタンパク質の動態や、染色体構造メンテナンス（SMC）タンパク質の動態を直接観察することで、それらの機能発現のメカニズムの理解を深める。特に、SMタンパク質では、分子のドメイン構造を識別しながらDNAと結合している様子や、DNAとの結合様式がヌクレオチドに依存して変化することを見出すことに成功しているので、重点的に研究を推進し、分子の機能メカニズムの詳細に迫る。また、真核細胞表面を高速AFM観察し、膜表面のタンパク質や脂質の形状動態を高時間・高空間分解能で得ることで、それらの実体を明らかにすることを目指す。
