Outline of Research at the Start

がん細胞選択性を持たせ、がん細胞に導入し、外部磁場を印加することでがん細胞のみを加温する「磁気ハイパーサーミア」により、副作用のないがん治療の可能性を目指す。独自の製法により、磁気ナノ微粒子を作製し、薬剤や抗体などを担持することが可能な、高度な機能化を持つ微粒子を開発するとともに、それらを用いて温熱療法、磁気ハイパーサーミアを実現する手法を提案する。さらには開発した微粒子をMRIやCT、磁気イメージング(MPI)等にも活用し、診断gnostics)と同時に治療(Therapy)が可能な画期的な「セラノスティクス」の潮流を生みだすことを目的とする

Outline of Annual Research Achievements

独自の製法でスーパースピングラス(SSG)磁気ナノ微粒子を生成し、特に複素磁化率に注目しながら磁気的性質と熱エネルギー蓄積機構を明らかにするとともに、官能基や葉酸を修飾し、医療応用への可能を検討した。交流磁場中で磁気微粒子が発熱することを利用した、がん温熱療法へ応用するため、熱散逸と緩和による発熱メカニズムを分析した。今年度は特に以下の5項目に注目した。
1. 粒径が6-19 nmのZn0.2Fe2.8O4のSSG磁気ナノ微粒子を作製し、ネール緩和・ブラウン緩和による発熱機構を分離する試みを行った。その結果、比吸収率(SAR)を見ると、15 nm以下の試料ではネール緩和が支配的、19 nmではブラウン緩和が支配的であることが示唆された。
2.がん細胞に選択的に導入させるための1案として、グルコースの修飾に挑戦した。まずポリエチレングリコール(PEG)での包含を試みた。PEG修飾により、水中分散性は向上した。がん細胞への導入率にはわずかな優位性が観察できた。
3. コイルについては、従来のソレノイド型からより高磁場を得られるように、また、将来的には無理なくヒトへの適用が可能なように、Zero-Voltage-Switching回路を導入した。上昇温度は従来の3倍ほどになり、コイルの中心から離れた位置でも昇温が可能になることを確認した。
4.亜鉛(Zn)をフェライトにドープすることで、磁化が増大することを見出したが、放射光を用いた局所構造解析により、Znは100%Aサイトに配位することが明らかになった。
5. 磁気微粒子イメージング（Magnetic particle imaging, MPI)についての装置の構築とデータの分析を進めたMn-Zn ferriteにGdイオンをドープした系にて、第3高調波成分が明瞭に観測され、高い機能が期待されることがわかった。
