Outline of Research at the Start

睡眠は動物に普遍的に認められ心身の健康に必須である。しかし、睡眠制御の分子機構は明らかになっていない。研究代表者らは新しい睡眠制御分子としてリン酸化酵素SIK3を同定しSIK3が睡眠必要量を規定する分子機構を構成していることを明らかにした。SIK1およびSIK2も同様に役割を持つことを明らかにしつつある。本研究ではSIK3がその下流分子と協働して睡眠必要量を決める仕組みを最新の知見と技術を用いて明らかにする。

Outline of Annual Research Achievements

睡眠覚醒制御の実行回路を構成する神経細胞の働きを変化させる細胞内の仕組みは明らかではない。研究代表者らは順遺伝学的アプローチにより新しい睡眠制御分子、特に睡眠要求を調整する役割を持つ分子としてリン酸化酵素SIK3を同定した。Sleepy変異型のSIK3を発現すると睡眠が増大する。睡眠制御におけるSIK3の基質の同定が課題となっていた。睡眠の表現型から機能獲得型変異と想定されていたが、精製した蛋白質を用いるin vitro kinase アッセイでは機能獲得型の変化は認められなかった。培養細胞を用いたHDAC4を介した転写アッセイでは機能獲得型変化を示したことから、睡眠の変化もHDAC4を介したものと理解できる。HDAC4の睡眠における役割の一端を前年度に公表したが、さらに検討を続けておりClass II HDACの代償性や発現部位の違いも考慮し、発生期や末梢組織の影響を除外することで睡眠制御の分子機構が徐々に明らかになってきた。睡眠覚醒行動に密接な関係を持つ概日リズム行動についてもHDAC4が関与することを、HDAC4ヘテロ欠損マウスを用いることで示すことができた。HDAC4ヘテロ欠損マウスは恒暗条件での概日リズム長が野生型マウスよりも短縮していた。SIK3欠損とは逆向きの変化であり、SIK3がHDAC4をリン酸化することでHDAC4の核内への移行を阻害しHDAC4の転写抑制を制限することで統一的に理解できる。HDAC4ヘテロ欠損マウスは暗期初期の覚醒開始時間に野生型マウスとの差異を示さなかったが、明期側への前倒しは光のマスク効果のために検出できにくいためだろう。このように、HDAC4が睡眠及び概日リズムにおいて重要な役割を果たしていると示すことができた。
