Outline of Research at the Start

生命体は、これまでに人類が知り得てきた科学技術とは、その動作原理が根本的に異なっている。本研究では、生命体を非平衡開放系における階層的な自律的システムと捉え、実空間上で生命の動作特性を発現する実空間のモデル系を構築し、それにより、「要素還元的な分子論を中核とする生命科学」と、「コンピュータ空間でのモデル化に象徴される数理科学」の学問の流れを統合し、新しい学問の潮流を創出するとともに、医学や工学的な分野への応用展開も併せて図っていくことを目的としている。次のような課題を重点てきに追究する。1)ゲノムDNAの高次構造転移と遺伝子活性、2) 細胞の実空間モデリング、3) 細胞の３次元組織体自己生成。

Outline of Annual Research Achievements

実空間上に単純な実験モデルを構築するとともに、非平衡物理学や非線形科学に立脚した数理モデルと対比しながら、以下の3つの課題を中核に研究を総合的に推進した。
１）ゲノムDNAの高次構造転移と遺伝子活性に関して、溶液中での１分子の動的高次構造観測を活用し、高次構造と転写・発現・複製の活性との関連を明らかにすることを目指して研究を推進した。無細胞発現系を用いた実験により、一価カチオンの影響を定量的に調べた。その結果発現活性が、Rb>K>Cs>Na≒Li 、このような順番となることを明らかにした。ethanolなどのアルコールが発現活性を亢進させることを見出し、これが一分子鎖観察のデータを解析することにより求めた長鎖DNAの弾性の変化と相関していることを明らかにしている。抗がん剤Daunomycinなどが遺伝子活性とDNA高次構造変化に対して、特徴的な作用を示すことなども解明した。
２)細胞の実空間モデリングでは、水/水ミクロ相分離液滴を生成させるときに、微小管やキネシンを共存させると、多様な変形を示す細胞様の構造が自己組織的に創成されることを見出した。さらには、ガラス細管内で水/水相分離を起こすと、規則的に配列しサイズの揃った細胞サイズの液滴が生じることを明らかにしている。また、細胞の混雑環境が細胞顆粒の特異的な局在化を引き起こすことを、マクロな実空間のモデル（振動盤上の粒子粒子群の挙動）を用いて明らかにし、理論的なモデルも用い、現象の一般性を理論・実験両面化が解明してきている。
3)細胞の３次元組織体自己生成では、水/水ミクロ相分離液滴を用いた細胞モデル実験で、異種液滴が交互に配列するような、自己組織化構造が自発的に生成するといった、新たな実験系の創出ができている。がん細胞の全ゲノム解析により、活性な遺伝子群の時間発展の過程を明らかにするといった内容の研究も推進した。
